Novel phenylalanine dehydrogenases from Sporosarcina ureae and Bacillus sphaericus. Purification and characterization.
NAD+-dependent phenylalanine dehydrogenases were purified 1,500- and 1,600-fold, and crystallized from Sporosarcina ureae SCRC-R04 and Bacillus sphaericus SCRC-R79a, respectively. The purified enzymes were homogeneous as judged by disc gel electrophoresis. The enzyme from S. ureae has a molecular weight of 305,000, while that of B. sphaericus has a molecular weight of 340,000. Each is probably ...
متن کاملStructure of the regular surface layer of Sporosarcina ureae.
Optical diffraction and computer image processing of electron micrographs were employed to analyze the structure of the regular surface layer of Sporosarcina ureae at high resolution. Negatively stained preparations of regular surface layer fragments showed two types of tetragonal pattern, each having p4 symmetry in projection with a = 12.8 nm. Although the two patterns differed greatly in over...
متن کاملSurface arrays on the wall of Sporosarcina ureae.
Thin sections of the cell wall of Sporosarcina ureae revealed two structurally distinct layers: a continuous amorphous zone, approximately 15 nm thick, which was adjacent to the plasma membrane, and an overlying periodic zone, approximately 16 nm thick. Sequential Triton X-100 and lysozyme treatment of isolated walls produced small fragments of the outer regular structure which allowed high-res...

Bacteriol. 90:808-816. 1965.-Ultrathin sections of Sporosarcina ureae cells were studied by means of electron microscopy. The cell wall consists of several layers and is 340 A thick. The cytoplasm is of globular structure and includes ribos...
متن کاملAmino Acid Isomerization in the Production of L-phenylalanine from D-phenylalanine by Bacteria.
To establish an advantageous method for the production of l-amino acids, microbial isomerization of d- and dl-amino acids to l-amino acids was studied. Screening experiments on a number of microorganisms showed that cell suspensions of Pseudomonas fluorescens and P. miyamizu were capable of isomerizing d- and dl-phenylalanines to l-phenylalanine. Various conditions suitable for isomerization by...
